Thrombosis and Hemostasis: Evolving Concepts and Novel Therapeutic Strategies

This chapter critically reviews recent advances in the understanding of thrombotic and bleeding disorders, encompassing venous thromboembolism (VTE), thrombophilia, hemophilia, and platelet disorders, as well as the increasingly recognized role of immunothrombosis and thromboinflammation in systemic disease. It discusses the clinical evidence base for direct oral anticoagulants (DOACs), the development of novel hemostatic agents for hemophilia A and B including extended half-life factor products and non-factor therapies, and the molecular pathogenesis of heparin-induced thrombocytopenia (HIT) and vaccine-induced immune thrombocytopenia and thrombosis (VITT). Emerging antithrombotic targets, including factor XIa and PAR-4 inhibitors, are also examined.
